Your command is wrong, you need to pass full and correct path to SOFA file.

I've tried:
-af sofalizer=sofa=C:\Users\User_000\Downloads\ClubFritz6.sofa:type=freq:radius=1
-af sofalizer=sofa=C:/Users/human_000/Downloads/ClubFritz6.sofa:type=freq:radius=1
-af sofalizer=sofa=\Users\User_000\Downloads\ClubFritz6.sofa:type=freq:radius=1
-af sofalizer=sofa=ClubFritz6.sofa:type=freq:radius=1 <-- (This one with ClubFritz in the same directory as FFMpeg)
-af sofalizer=sofa=/Users/User_000/Downloads/ClubFritz6.sofa:type=freq:radius=1
-af sofalizer=sofa="/Users/User_000/Downloads/ClubFritz6.sofa":type=freq:radius=1

All give me errors like

I tried put full path in thousand ways. No success! It simply doesn't work. Don't know, what is wrong.

I put ClubFritz6.sofa in the same directory and also to the root of drive D I am working on.

d:\Media\Movies\__Temp>ffmpeg.exe -i pokus2.mkv -af sofalizer=sofa=\ClubFritz6.sofa output.mkv
d:\Media\Movies\__Temp>ffmpeg.exe -i pokus2.mkv -af sofalizer=sofa=/ClubFritz6.sofa output.mkv
d:\Media\Movies\__Temp>ffmpeg.exe -i pokus2.mkv -af sofalizer=sofa=ClubFritz6.sofa output.mkv

d:\Media\Movies\__Temp>ffmpeg.exe -i pokus2.mkv -af sofalizer=sofa="\ClubFritz6.sofa" output.mkv
d:\Media\Movies\__Temp>ffmpeg.exe -i pokus2.mkv -af sofalizer=sofa="/ClubFritz6.sofa" output.mkv
d:\Media\Movies\__Temp>ffmpeg.exe -i pokus2.mkv -af sofalizer=sofa="ClubFritz6.sofa" output.mkv

All the similar output:

[Parsed_sofalizer_0 @ 000000000016bd00] Can't find SOFA-file 'ClubFritz6.sofa'
[Parsed_sofalizer_0 @ 000000000016bd00] Error while loading SOFA file: 'ClubFritz6.sofa'
[Parsed_sofalizer_0 @ 000000000016bd00] No valid SOFA file could be loaded. Please specify valid SOFA file.
[AVFilterGraph @ 0000000000167500] Error initializing filter 'sofalizer' with args 'sofa=ClubFritz6.sofa'
Error opening filters!

What the hell I am doing wrong? :-(

I changed filter to depend on libmysofa library in latest master, and also plan to add native filter with slightly complex syntax which would have same functionality.

So you are free to fetch latest ffmpeg and compile and try.

@richardpl - are you refer to lastly added headphone filter https://ffmpeg.org/ffmpeg-filters.html#headphone ?
Somehow documentation may be confusing - basically new filter is multichannel convolver where convolution coefficients must be provided trough separate list of wav files. Using sofalizer can be easier but it doesn't work - it will be nice to call headphone with for example filelist where separate wav's can be specified or use multichannel wav file.
Above comments are valid if this native filter mentioned by you is headphone. If not then sorry for bothering.

Well... Zeranoe builds obviously lack this filter (not sure why).
